OBJECTIVES: This study aimed to synthesize and characterize mesoporous zinc oxide nanoparticles (ZnO NPs) and also to evaluate the cytotoxicity of mesoporous ZnO NPs on L929 mouse fibroblast cell lines using 3-(4,5- dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide (MTT) assay. MATERIALS AND METHODS: The synthesized mesoporous ZnO NPs were extensively characterized using X-ray diffraction analysis (XRD), transmission electron microscopy (TEM), Brunauer-Emmett-Teller (BET) analysis, field emission scanning electron microscopy (FESEM), energy dispersive X-ray spectra (EDAX), Fourier-transform infrared spectroscopy (FTIR), and dynamic light scattering (DLS). The cytotoxicity of mesoporous ZnO NPs was assessed by MTT assay. The study groups for cytotoxicity assay were normal saline, 0.1% calcined mesoporous ZnO NP solution, 1% calcined mesoporous ZnO NP solution, 0.1% noncalcined mesoporous ZnO NP solution, 1% noncalcined mesoporous ZnO NP solution, 0.1% ZnO NP solution, 1% ZnO NP solution, 2% chlorhexidine, and phosphate-buffered saline (PBS). The percentages of mean ± standard deviation of viable cells were analyzed. RESULTS: Characterization of mesoporous ZnO NPs revealed that all the particles were in a more or less spherical shape with a wide particle size distribution of 70-100 nm. TEM image showed the uniformed and aggregated ZnO NPs with a typical size of 10-15 nm. BET analysis showed a mesoporous structure for the prepared mesoporous ZnO NPs. According to the MTT assay, chlorhexidine had the lowest cell viability percentage. Cell viability percentages of 0.1% mesoporous ZnO NP solutions (calcined and noncalcined) were statistically, significantly higher than 0.1% ZnO NP solution (p < .05). Cell viability percentages of 0.1% calcined and noncalcined mesoporous ZnO NP solutions and 0.1% ZnO NP solution were statistically, significantly higher than the 1% solutions (p < .05). CONCLUSION: Mesoporous ZnO NPs exhibited less cytotoxicity against L929 mouse fibroblast cell lines compared to CHX and ZnO NPs, hence are safe to use.

It has been shown that irradiation can cause structural changes in dentin that may reduce the bond strength of adhesives to dentin. Applying cross-linking or antioxidant agents may help reverse this detrimental effect and improve adhesion to dentin. This in vitro study aimed to evaluate the effects of epigallocatechin-3-gallate (EGCG) pretreatment and the time of adhesive bonding (24 hours vs 1 month) on the shear bond strength (SBS) of All-Bond Universal (ABU) to irradiated dentin using etch-and-rinse (ER) and self-etching (SE) modes. Flat dentin surfaces prepared from 96 extracted intact human molars were divided into 8 groups (n = 12) and bonded with ABU. In the control (CO) groups (CO/ER and CO/SE), bonding was performed on nonirradiated dentin; in the irradiated (IR) groups (IR/ER and IR/SE), bonding was performed on irradiated dentin; in the irradiated pretreated groups (IR/EGCG/ER and IR/EGCG/SE), irradiated dentin received a 0.1% EGCG pretreatment before bonding; and in the irradiated delayed bonding (DL) groups (IR/DL/ER and IR/DL/SE), bonding on irradiated dentin was performed 1 month after completion of radiotherapy. The irradiation protocol consisted of a total dose of 60 Gy with 2-Gy exposure applied 5 days per week for a period of 6 weeks. After bonding procedures were completed, the specimens were stored in 100% humidity at 37°C for 24 hours and then the SBS was tested in a universal testing machine. Data were analyzed using 1-way analysis of variance and Tukey tests. There was a statistically significant difference among the 8 groups (P < 0.001). Irradiation diminished the SBS in the IR/ER and IR/SE groups compared with their controls (P < 0.001). Pretreatment with EGCG significantly increased the SBS in the IR/EGCG/ER group only (P < 0.001). The difference between the IR/ER and IR/DL/ER groups was not statistically significant, and the difference between the IR/SE and IR/DL/SE groups was marginally significant (P = 0.056). Pretreatment with EGCG after acid etching restored the SBS of ABU to irradiated dentin, resulting in an adhesive performance equivalent to that observed with nonirradiated dentin. A 1-month delay between irradiation and bonding did not improve the SBS.

OBJECTIVE: This study evaluated the effect of two natural antioxidants on the compromised bond strength of a resin-modified glass ionomer (RMGI) to the sodium hypochlorite (NaOCl)-affected pulp chamber dentin. METHODS: Forty-two sound third molars were split into halves. The exposed pulp chamber dentin was ground to provide the flat dentin surfaces and divided into seven groups (n = 12), according to the solutions used for immersion: (1) Control, distilled water; (2) NaOCl, 5.25% NaOCl for 20 min; (3) NaOCl/Ethylenediaminetetraacetic acid (EDTA); 5.25% NaOCl for 20 min + 17% EDTA for 1 min; (4) NaOCl/TA, 5.25% NaOCl + 10% tanic acid (TA) for 5 min; (5) NaOCl/EDTA/TA, 5.25% NaOCl + 17% EDTA + 10% TA for 5 min; (6) NaOCl/PA, 5.25% NaOCl+ 10% proanthocyanidin for 5 min; and (7) NaOCl/EDTA/PA, 5.25% NaOCl+ 17% EDTA + 10% PA for 5 min. The RMGI was bonded on the treated dentin using a Tygon tube. After 24 h of storage, microshear bond strength (µSBS) was tested. Data in MPa were submitted to one-way analysis of variance and Tamhane test. RESULTS: NaOCl significantly decreased the µSBS; NaOCl/EDTA and NaOCl/TA significantly increased the µSBS, higher than the control group (p < .05); and in the NaOCl/EDTA/TA group, the increased bond strength was at the level of the control group (p > .05). NaOCl/PA and NaOCl/EDTA/PA and NaOCl groups had comparable µSBS. CONCLUSION: TA could be suggested to provide effective bonding of RMGI and immediate sealing of the pulp chamber dentin after NaOCl irrigation.

OBJECTIVE: Given the importance of preserving caries-affected dentin (CAD) in conservative dentistry, the shear bond strength (SBS) of different resin cements to CAD has been investigated. Here, we aimed to compare the SBS and remineralizing effect of a calcium silicate (TheraCem) and conventional self-adhesive cement (Panavia SA) on the SBS of CAD. MATERIALS AND METHODS: Forty-eight extracted third molars (24 sound and 24 CAD) were used. In each group, 12 teeth were prepared for bonding to TheraCem or Panavia SA. After removal of the enamel and caries, resin composite cylinders were luted on the prepared dentin. After 28 days of storage in the artificial saliva, SBS was measured and the failure mode analysis was investigated. The images of fractured sections were analyzed using scanning electron microscopy and energy-dispersive X-ray to evaluate the Ca/P weight ratio. RESULTS: SBS of CAD and sound dentin was not different when cemented with TheraCem (9.56 ± 4.51 vs. 9.17 ± 2.76, p = .806), but the CAD showed significantly lower SBS to Panavia SA (9.4 ± 2.36 vs. 7.39 ± 2.18, p = .015). The Ca/P ratio in CAD was significantly higher when bonded to both TheraCem and Panavia-SA than that of the controls (p = .001); however, this ratio was not different for those bonded to TheraCem compared to Panavia SA. CONCLUSIONS: Based on our results, TheraCem as a calcium silicate cement shows better SBS to attach the restoration to CAD as compared to Panavia SA. Obliteration and mineralization of the dentinal tubules in TheraCem were also higher than in Panavia SA. However, their ability to improve the amount of the Ca/P ratio in CAD was similar.

Introduction: Considering the recent trend to use mineral trioxide aggregate (MTA) and Biodentine and resin cements, more conservative approaches concurrent with adequate bond strength have always been requested. The present study aimed to evaluate the effect of pretreatment with Er:YAG laser etching versus acid-etching for 5 and 15 seconds on the micro shear bond strength of self-adhesive resin cement (SRC) to MTA and Biodentine. Materials and Methods: Forty-eight samples of each cement (MTA and Biodentine) were prepared and distributed into four groups based on surface pretreatment: 1) control, no treatment; 2) Er:YAG laser etching with energy of 60 mJ; 3) 5-second acid-etching; 4) 15-second acid-etching. All specimens were cemented using SRC. Microshear bond strengths were tested following 24-hour water storage. Debonded specimens were examined and surface topography was assessed using an atomic force machine (AFM). Data analysis was performed using the two-way ANOVA and Tukey multiple comparisons test. Results: The three testing groups of laser etch and 5-s and 15-s acid-etch demonstrated a significantly higher SBS than the control group (P < 0.05) with negligible differences among them (P > 0.05). Furthermore, Biodentine showed better adhesive bonding than MTA in all groups. Conclusion: Laser etching of 60 mJ and 5-s acid-etching were as beneficial as 15-s acid-etching in terms of bond strength of SRC to MTA and Bodentine.

BACKGROUND: Coronal restoration of endodontically treated teeth (ETT) with mesio-occluso-distal (MOD) cavities is of a great importance in long-term success of the treatment. This study evaluated the effect of fiber reinforcement on the fracture resistance (FR) of ETT restored with flowable or paste bulk (PB)-fill composite resin compared to conventional composite (CC) resin. MATERIALS AND METHODS: In this in vitro experimental study, eighty maxillary premolars were divided into eight groups (n = 10). The first group was left intact (G1) and the other groups received MOD cavities along with endodontic treatment. G2: Remained unrestored while the other experimental groups were restored with three types of composite resin with or without fiber insertion. G3: CC resin, G4: PB fill, G5: Flowable bulk fill (FB). G6: Fiber + CC, G7: Fiber + PB, and G8: Fiber + FB. FR was tested at crosshead speed of 1 mm/min and recorded in Newton. Data were analyzed using one-way analysis of variance and Tukey's tests at significance level of P < 0.05. RESULTS: G1 and G2 revealed the highest and the lowest FR, respectively. The mean FR of the testing groups in Newton was as follows: G1 = 1204.8 A, G2 = 352.1 C, G3 = 579.6 BD, G4 = 596.7 BD, G5 = 624.9 BDE, G6 = 858.3 E, G7 = 529.6 CB, and G8 = 802.5DE. Different uppercase letters indicate the significant difference between the groups. CONCLUSION: The effect of fiber insertion on FR depended on the type of composite resin; the highest reinforcing effect was obtained in the CC resin + fiber, followed by bulk-fill flowable + fiber, and flowable bulk (FB)-fill composite resin. The strength of the former was significantly higher than the conventional and PB fill with and without fiber.

PURPOSE: To evaluate the proportion of microleakage (PM), shear bond strength (SBS), and the fissure sealant (FS) interface by scanning electron microscopy (SEM) in three kinds of FS when the enamel surfaces were contaminated with saliva. METHODS: 198 sound third molar teeth were randomly divided into three pretreatment condition groups (n = 66): dry, saliva contamination removed by cotton pellet, or saliva removed by air-drying. A resin-based FS (Clinpro™), amorphous calcium phosphate-containing FS (Aegis®), or glass ionomer-based FS (Fuji Triage®) was applied on the treated enamel, and PM and SBS were assessed. Two specimens from each group were observed with SEM. p values < 0.05 was considered statistically significant. RESULTS: Glass ionomer-based FS showed the highest PM in all three surface conditions (p values < 0.05 were considered statistically significant). No significant difference in PM was observed between resin-based FS and amorphous calcium phosphate-containing FS (p > 0.05). Resin-based FS showed significantly greater SBS in all three surface conditions compared to glass ionomer-based FS. SEM observations showed that saliva contamination led to gaps at the enamel-sealant interface. CONCLUSION: Neither cotton pellet-drying nor air-drying effectively removed saliva from the contaminated enamel surface. Glass ionomer-based FS showed the highest PM and the lowest SBS in contaminated and noncontaminated conditions. The highest SBS was obtained with resin-based FS.

AIM: This study aimed to evaluate the effects of various restorative techniques on the fracture resistance of pulpotomized premolars with mesioocclusodistal (MOD) cavities treated with mineral trioxide aggregate (MTA) or calcium enriched mixture (CEM) cement. MATERIALS AND METHODS: One hundred and eight sound extracted maxillary premolars were randomly assigned to nine experimental groups (n = 12). The teeth in group 1 did not receive any preparation. Class II MOD cavities were prepared in the other experimental groups. In groups 2, 4, 6, and 8, tooth-colored MTA was used for pulpotomy. In groups 3, 5, 7, and 9, CEM cement was used for pulpotomy. Groups 2 and 3 were left unrestored. Groups 4 and 5 were restored with amalgam. Groups 6 and 7 were restored with a conventional composite resin, and groups 8 and 9 were restored with bulk-fill giomers. Fracture resistance was measured, the fracture pattern of each specimen was assessed, and the results were statistically analyzed. RESULTS: The fracture resistance of group 1 was significantly higher than those of the other groups (p < 0.05). The fracture resistance of group 2 (MTA + amalgam) was statistically lower than those of all experimental groups (p values < 0.05) except groups 3, 4, and 5 (p values > 0.05). No statistically significant differences were found between the groups restored with amalgam, conventional composite resin, and bulk-fill giomer (groups 4, 5, 6, 7, 8, and 9) (p values < 0.05). The highest rate of mode 1 fracture (restorable fracture) was observed in group 1 followed by groups 8 and 9. CONCLUSION: No significant differences were found among the fracture resistances of the restored teeth using various restorative techniques. Bulk-fill giomers followed by conventional composite resin were better able to prevent unfavorable fractures compared to amalgam. Therefore, they seem to be more reliable for the restoration of pulpotomized teeth with MOD cavities.

OBJECTIVES: This study aimed to investigate the influence of the operator's experience on the microleakage of two universal adhesives applied in self-etch (SE) and etch-and-rinse (E&R) modes. MATERIALS AND METHODS: Two class V cavities were prepared on the buccal and lingual surfaces of 112 caries-free extracted human molars and were divided into eight groups based on the combination of the factors "adhesive system" (Tetric N-Bond Universal Adhesive [TNU]; Scotchbond Universal Adhesive [SBU]), "adhesive strategy" (SE or E&R), and "the operators performing the procedures" (specialists in operative dentistry or intern students). After 24 h of water storage and thermocycling, the marginal microleakage was evaluated using the dye penetration technique and the data were analyzed. RESULTS: In the SE mode, microleakage at the enamel margin for both adhesives was higher in the student group than in the specialist group (p values < 0.05). The operator's skill did not affect the microleakage scores of both adhesives applied in the E&R mode at the dentin and enamel margins or in the SE mode at the dentin margins (p > 0.05). The microleakage score was significantly higher at the dentin margins than the enamel margins for both universal adhesives applied in the E&R mode (p values <0.05). CONCLUSION: The microleakage values of the universal adhesives applied in the SE mode at the enamel margins were affected by the operator's skill. However, the operator's experience did not affect the microleakage scores of the two universal adhesives applied in the E&R mode at the enamel and dentin margins or in the SE mode at the dentin margins.

This study aimed to compare the effects of different combinations of adhesive bases and restorative materials on the fracture strength and mode of maxillary premolars with mesio-occlusodistal (MOD) cavities after mineral trioxide aggregate (MTA) pulpotomy. Ninety-six extracted human maxillary premolars were divided into 8 groups (n = 12). Group 1 (negative control) consisted of intact teeth. In the other teeth, MOD and endodontic access cavities were prepared, and a layer of MTA was placed. Group 2 was left unrestored as the positive control. Group 3 was restored with a glass ionomer cement (GIC) base and amalgam. The remaining groups were restored with a microhybrid composite after application of different bases: 4, resin-modified GIC (RMGIC); 5, zirconia-reinforced GIC (ZRGIC); 6, self-adhesive flowable composite (SAFC); 7, self-adhesive resin cement (SARC); and 8, short fiber-reinforced composite (SFRC). After fracture strength testing via continuous compressive axial loading, the fracture mode was classified as restorable or unrestorable. Data were analyzed using 1-way analysis of variance and post hoc Tamhane tests (P < 0.05). The fracture strength of the negative control group was significantly higher than that of all other groups (P < 0.001). The fracture strengths of groups 2 and 3 were not significantly different (P > 0.05) from each other but were significantly lower (P = 0.002) than those of all composite-restored groups. Group 8 showed a significantly greater fracture strength than group 4 (P < 0.001). Unlike GIC/amalgam, all of the base/composite restoration groups partly restored the strength of pulpotomized premolars. Although their fracture strengths were statistically similar, the fracture modes were more favorable in groups with SAFC or SARC bases than in groups with RMGIC or ZRGIC bases. The SFRC/composite specimens revealed advantages in both fracture strength and fracture mode compared to RMGIC/composite specimens.

This in vitro study aimed to evaluate the influence of endodontic irrigation solutions and resin sealing of the dentin surfaces of the cavity before irrigation on the fracture strength (FS) of maxillary premolars. Seventy-two human maxillary premolars were randomly assigned to 6 groups (n = 12). Group 1 consisted of intact teeth that served as negative controls. In groups 2 to 6, root canal treatment was performed after mesio-occlusal preparation. In group 6, the dentin surfaces of the mesio-occlusal cavity were sealed with a self-adhesive flowable composite resin before instrumentation and irrigation of the canal. The volume and contact time of the irrigation solutions used during endodontic treatment of groups 2, 4, 5, and 6 were standardized as follows: 2 mL of 5.25% sodium hypochlorite (NaOCl) for 1 minute after each file, 5 mL of 17% ethylenediaminetetraacetic acid (EDTA) for 3 minutes after instrumentation was completed, and 5 mL of 5.25% NaOCl for 1 minute as the final irrigation. In group 3, the irrigation solution was normal saline solution only. After root canal obturation, in group 2, the cavities were left unrestored as a positive control. In groups 3 (saline-rinsed), 4 (conventionally restored), and 6 (presealed), the restorations were carried out with an etch-and-rinse adhesive and composite resin immediately after root canal obturations. In group 5 (delay-restored), the restorations were carried out with an etch-and-rinse adhesive and composite resin after a 1-week delay. After thermomechanical cycling, the teeth were subjected to FS testing under continuous compressive force. Data were analyzed with 1-way analysis of variance and Tukey tests. The unrestored group, which demonstrated the lowest FS, did not differ significantly from the conventionally restored group; the FS of both groups was significantly lower than that in the other groups (P ≤ 0.001). The presealed group exhibited the highest FS (P ≤ 0.02). The FS values of the saline and delay-restored groups were statistically comparable. In the present study, NaOCl/EDTA irrigation had an adverse effect on the FS. Presealing of the dentin surfaces of the cavity with self-adhesive flowable composite resin significantly increased the FS of aged composite resin-restored premolars.

AIM: Simulated pulpal pressure (PP) has been shown to affect the bond strength and nanoleakage of different adhesives at dentin interfaces but the effect of simulated PP on polymerization of adhesives has not been studied yet. Furthermore, it has been proposed that strong and mild self-etch adhesives have different polymerization behaviors. This study aimed to evaluate the effect of simulated PP on polymerization of two self-etch adhesives, Adper Prompt L-Pop (APLP) and Adper Easy Bond (AEB), by means of the Knoop hardness test. MATERIALS AND METHODS: Sixty caries-free human molars were used to prepare deep dentin specimens with a mean remaining dentin thickness of 0.9 mm. The specimens were bonded in the absence or presence of PP. The specimens were assigned to four equal groups (n = 15) as follows: AEB/-PP, APLP/-PP, AEB/+PP, and APLP/+PP. Bonding procedures were completed; then the specimens' hardness was measured with the Knoop test. Data were analyzed with two-way ANOVA and the t test. RESULTS: In the absence of PP, the hardness of AEB was significantly higher than that of APLP (p < 0.001). In contrast, when PP was simulated, the hardness of APLP was higher than that of AEB (p = 0.002). The hardness of AEB was not influenced by the presence of PP (p = 0.153). Simulation of PP resulted in a significant improvement in the hardness of APLP (p < 0.001). CONCLUSION: The polymerization degree of strong self-etch adhesive was lower compared to mild self-etch adhesive. In the presence of hydrostatic PP, the polymerization degree of strong self-etch adhesive was higher than mild self-etch adhesive. CLINICAL SIGNIFICANCE: Dentin moisture caused by positive PP might improve polymerization of strong self-etch adhesives.

RESUMO

This study was conducted to evaluate whether antibacterial pretreatment irrigation with silver nanoparticles (SNPs) and silver diamine fluoride (SDF) has any effect on bond strength of fiber posts cemented with three types of resin cements in root canal space. Fifty-four endodontically treated maxillary central incisor roots were prepared for fiber post (FRC Postec Plus NO.3, Ivoclar Vivadent) cementation and divided into nine groups in terms of three cement types and two pretreatments with silver antibacterial agents. The cements were as follows: an etch-and-rinse cement (ER, Excite DSC/Variolink N), a self-etch cement (SE, ED Primer/Panavia F2.0), and a self-adhesive cement (SA, Panavia SA Luting Plus). For each cement, the control group was with no treatment and two experimental groups were with SNPs and SDF treatments that were used after acid-etching for ER cement and after EDTA treatment for SE and SA cements. After fiber post cementation, each bonded root was horizontally sectioned into 1-mm thickness microslices to create two slices for each root region (apical, middle and coronal) and underwent push-out bond strength (PBS) test. Data in MPa were analyzed with two-way ANOVA and Tukey test (p=0.05). The interaction of the pretreatment type and cement type was significant (p<0.001). SNPs and SDF significantly increased PBS with ER cement (p≤0.04). This positive effect was also marginally significant for SDF with SE cement (p=0.049). For SA cement, SNPs showed a significant positive effect, but SDF had a significant adverse effect on PBS (p<0.001). The effect of pretreatment with silver antibacterial agents prior to adhesive cementation of fiber posts depends on the resin cement used. Contrary to SNPs with beneficial or no significant effect on bonding for all cements, SDF exhibited a deleterious effect with self-adhesive cement.

AIM: This study was conducted to evaluate the effects of three nanoparticle solutions used as dentin pretreatments on the microshear bond strength (µSBS) of a conventional glass ionomer cement (GIC) to dentin. MATERIALS AND METHODS: Ninety intact human molars were used after sectioning their occlusal surfaces to expose flat dentin surfaces. The specimens were randomly assigned to nine groups (n = 10). Group A was the control group (without using the cavity disinfectant). In groups B, C, D, and E, the prepared dentin surfaces were treated with 1 cc 2% chlorhexidine (CHX), 0.1% silver nanoparticle (SNP), 0.1% titanium dioxide nanoparticle (TNP), and 0.1% zinc oxide nanoparticle (ZNP) solutions for 1 minute, respectively, before applying the conditioner. CHX, SNPs, TNPs, and ZNPs were applied for 1 minute after applying the conditioner in groups F, G, H, and I, respectively. The specimens were restored with a conventional GIC and underwent µSBS testing after 24 hours. The data were analyzed using the one-way analysis of variance and Tukey's test (p=0.05). RESULTS: The applications of the nanoparticles (SNP, TNP, and ZNP) after the conditioner were associated with significantly greater µSBS values compared to that of the control group (p values < 0.05). Significantly higher µSBS values were observed when TNP or ZNP was applied after the conditioner compared to their applications before the conditioner (p values < 0.05). The highest µSBS values were observed when TNP was applied after the conditioner. CONCLUSION: Dentin pretreatment with the nanoparticles after applying the conditioner enhanced the bond strength of the GIC to dentin compared with the control group. The best results were obtained for the TNPs applied after the conditioner.

Resumo Este estudo foi conduzido para avaliar se a irrigação de pré-tratamento antibacteriano com nanopartículas de prata (SNPs) e fluoreto de diamina de prata (SDF) tem algum efeito na resistência de união de pinos de fibra cimentados com três tipos de cimentos resinosos no espaço do canal radicular. Cinquenta e quatro raízes de incisivos centrais superiores tratadas endodonticamente foram preparadas para cimentação de pino de fibra (FRC Postec Plus NO.3, Ivoclar Vivadent) e divididas em nove grupos em termos de três tipos de cimento e dois pré-tratamentos com agentes antibacterianos de prata. Os cimentos foram os seguintes: um cimento etch-and-rinse (ER, Excite DSC / Variolink N), um cimento autocondicionante (SE, ED Primer / Panavia F2.0) e um cimento autoadesivo (SA, Panavia SA Luting Plus). Para cada cimento, o grupo controle foi sem tratamento e dois grupos experimentais foram com SNPs e SDF tratamentos que foram usados após o condicionamento ácido para o cimento ER e após o tratamento com EDTA para os cimentos SE e SA. Após a cimentação do pilar de fibra, cada raiz ligada foi seccionada horizontalmente em microclimas de 1 mm de espessura para criar duas fatias para cada região da raiz (apical, média e coronal) e submetida ao teste de resistência de união por push-out (PBS). Os dados em MPa foram analisados ​​com ANOVA de dois fatores e teste de Tukey (p = 0,05). A interação do tipo de pré-tratamento e tipo de cimento foi significativa (p<0,001). SNPs e SDF aumentaram significativamente o PBS com cimento ER (p≤0,04). Este efeito positivo também foi marginalmente significativo para SDF com cimento SE (p = 0,049). AIM: This study was conducted to evaluate the effect of three nanoparticle solutions used as final root canal irrigants on the fracture resistance of endodontically treated roots. MATERIALS AND METHODS: Sixty intact single-rooted premolar teeth were used after removing the crowns below the cementum-enamel junction to standardize the length of the remaining roots to 13 mm. After instrumenting the roots using ProTaper Universal rotary instruments up to size F4, the roots were randomly divided into six groups (n=10) according to the final irrigating solutions: Group 1: normal saline; Group 2: 2% chlorhexidine (CHX); Group 3: 17% EDTA+2.5% NaOCl; Group 4: 17% EDTA+0.1% silver nanoparticle (SNP) solution; Group 5: 17% EDTA+0.1% titanium dioxide nanoparticle (TNP) solution; Group 6: 17% EDTA+0.1% zinc oxide nanoparticle (ZNP) solution. After filling the root canals with gutta-percha and AH Plus sealer, the fracture resistance (FR) values were measured. The data were subjected to the one-way analysis of variance and the Tamhane post hoc test (p=0.05). RESULTS: EDTA + NaOCl had the lowest FR value, followed by normal saline and CHX (p values < 0.05). No significant difference was found between the FR of roots irrigated with CHX and normal saline (p > 0.05). The application of nanoparticles (SNP, TNP, and ZNP) was associated with a significantly greater FR than that of other irrigation solutions (p values < 0.05). CONCLUSION: The final irrigation of root canals with nanoparticles enhanced the fracture resistance of the endodontically treated roots. The lowest FR value was observed for NaOCl.

INTRODUCTION: The stem cell portion of the dental pulp derived cultures (DPSCs) showed a higher resistance to cytotoxic effect of restorative dental materials compared to pulpal fibroblasts (DPFs). Here, we aimed to compare the expression of some drug resistant genes between these cells. METHODS AND MATERIALS: To separate DPSCs from DPFs, we used magnetic cell sorting technique based on CD146 expression. To assess the stem cell properties, the positive and negative portions underwent colony forming assays and were induced to be differentiated into the adipocytes, osteoblasts, hepatocytes, and neural cells. Cell surface antigen panels were checked using immune fluorescence and flow-cytometry techniques. The mRNA expression of 14 ABC transporters including ABCA2, ABCB1, ABCB11, ABCC1, ABCC2, ABCC3, ABCC4, ABCC5-2, ABCC5-4,ABCC5-13, ABCC6, ABCC10, ABCC11, and ABCG2 genes was assessed, using quantitative RT-PCR technique. RESULTS: Only the CD146 positive portion could be differentiated into the desired fates, and they formed higher colonies (16.7 ± 3.32 vs. 1.7 ± 1.67, p < .001). The cell surface antigen panels were the same, except for CD146 and STRO-1 markers which were expressed only in the positive portion. Among the ABC transporter genes studied, the positive portion showed a higher expression (approximately two-fold) of ABCA2, ABCC5-13, and ABCC5-2 genes. CONCLUSION: Dental pulp stem cells which can be separated from dental pulp fibroblasts based on CD146 expression, express higher levels of some drug resistance genes which probably accounts for their features of more resistance to cytotoxic effects of some dental materials. This needs to be more validated in future.

BACKGROUND: Immediate dentin sealing (IDS) with proanthocyanidin (PA) could be used before cementation with a self-adhesive (SA) cement. The aim of this study was to assess the effect of PA treatment on acid-etched dentin before adhesive application, in IDS and delayed dentin sealing (DDS), on the strengthening property of SA-cemented composite resin inlay in premolars. MATERIAL AND METHODS: Eighty-four maxillary premolars were divided into 7 groups (n=12): 1) (Intact) Sound teeth served as controls; groups 2-7) After cavity preparation and fabrication of composite resin inlay, temporary inlays were made and cemented. After one week, the inlays were removed and composite inlays were luted with a self-adhesive resin cement as follows: 2) (SA) Without dentin- pretreatment; 3) (DDS) Etch-and-rinse adhesive before the cementation; 4) (DDS/PA) PA treatment of acid-etched dentin before the adhesive, followed by the cementation; 5) (Etch/PA) PA treatment of acid-etched dentin before the cementation; 6 and 7) (IDS and IDS/PA) Application of IDS without or with PA treatment, respectively, one week before the cementation. After thermo-mechanical aging, fracture resistance (FR) was tested. Data were analyzed using one-way ANOVA and Tamhane tests (α=0.05). RESULTS: There was a significant difference between the study groups (P<0.001). The IDS and IDS/PA groups yielded significantly higher FR compared to the SA group (P ≤ 0.003), but the DDS, DDS/PA and Etch/PA groups did not differ from the SA group (P>0.05). The effect of PA on FR in the IDS and DDS techniques was not significant. CONCLUSIONS: IDS with or without PA treatment considerably improved the strength of premolars with self-adhesive-cemented inlay, while the value of only IDS with PA treatment reached the level of the sound teeth. BACKGROUND: This study was undertaken to investigate the effect of intraradicular dentin pretreatment with silver nanoparticles (SNPs), zinc oxide nanoparticles (ZNPs), and titanium oxide nanoparticles (TNPs) on the push-out bond strength (PBS) of fiber posts to root dentin using two types of resin cements. MATERIAL AND METHODS: Eighty single-rooted human premolar roots were randomly divided into eight groups after endodontic treatment and post space preparation, according to the type of intraradicular dentin pretreatment with different nanoparticle solutions (n=20). The groups included no pretreatment (control) and pretreatments with SNPs, ZNPs, and TNPs. Each group was divided into 2 subgroups (n=10) according to cementation: Excite DSC/Variolink II and ED Primer II/Panavia F2.0. The PBS values were measured in different areas of the post space. The data were subjected to the three-way analysis of variance and Tukey tests (p=0.05). RESULTS: The SNP-pretreated groups showed significantly higher PBS values than the other groups. No significant differences in PBS were noted among the control, ZNP-pretreated, and TNP-pretreated groups. There was no significant difference in the PBS of the fiber posts with respect to cement type. The PBS was significantly lower in the apical region than in the other two regions. There was no statistically significant difference between the PBSs of the cervical and middle thirds. CONCLUSIONS: Intraradicular dentin pretreatment with SNPs, TNPs, and ZNPs did not interfere with the PBS of the fiber posts. The best result was obtained for the SNP-pretreated groups for both types of cement. Also, the lowest PBS was found in the apical third of the root canal spaces. OBJECTIVE: To evaluate the effect of Er:YAG etching on topography, microstructure, compressive strength, and shear bond strength (SBS) of All-Bond Universal adhesive to mineral trioxide aggregate-Angelus (AMTA) and Biodentine (BD). METHODS AND MATERIALS: Sixty cylindrical specimens of each cement (AMTA and BD) in five groups were prepared and stored for 72 h. The control groups were non-etched, and four other groups were acid-etched and laser-etched with a pulse energy of 60, 80, or 100 mJ, followed by compressive strength testing. Surface micromorphology and topography were evaluated. Similar groups were bonded using All-Bond Universal with self-etch and etch-and-rinse (acid-etch) approaches, and laser-etch 60, 80, and 100 mJ, and SBS was tested. Data were analyzed using two-way and one-way ANOVA and the Bonferroni post hoc tests (α = 0.05). RESULTS: BD had a significantly higher compressive strength and SBS (except for laser-etch 100) compared to AMTA, regardless of the etching method (p < 0.001). Acid etching and laser etching 100 of both cements and laser etching 80 of BD alone produced a significantly lower compressive strength than that for the other groups. Contrary to BD, for AMTA, all the treatments significantly increased SBS compared to that of the self-etch group. CONCLUSIONS: Etching of AMTA was needed for stronger bonding; laser etching with 60 or 80 mJ without compromising compressive strength was recommended. Etching not only did not improve bonding ability of BD, but it also negatively affected the strength of BD. CLINICAL RELEVANCE: To achieve successful combined calcium silicate cement-resin material restoration, an adequate bond between the materials is mandatory. This might be provided with the ultramild adhesive through laser etching without compromising compressive strength, depending on cement composition and laser energy level used.
